Chloroquine mechanism of drug action and resistance in plasmodium falciparum

Chloroquine has been extensively used in mass drug administrations, which may have contributed to the emergence and spread of resistance. It is recommended to check if chloroquine is still effective in the region prior to using it.

The emergence of chloroquine resistance has been associated with a dramatic increase in malaria mortality among inhabitants of some endemic regions. The rationale for chemoprophylaxis is weakening as multiple‐drug resistance develops against well‐tolerated drugs. Plasmodium falciparum drug‐resistant malaria originates from chromosome. Drug-resistant P. falciparum. Chloroquine-resistant P. falciparum first developed independently in three to four areas in Southeast Asia, Oceania, and South America in the late 1950s and early 1960s. Since then, chloroquine resistance has spread to nearly all areas of the world where falciparum malaria is transmitted. The emergence of Plasmodium falciparum resistance to anti-malarial drugs has thwarted malaria control efforts and remains a major obstacle to malaria elimination throughout the world. Chloroquine was one of the first drugs to be used on a wide scale for the treatment of malaria. Chloroquine resistance emerged independently in different geographic regions.

The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.

The emergence of chloroquine resistance has been associated with a dramatic increase in malaria mortality among inhabitants of some endemic regions. The rationale for chemoprophylaxis is weakening as multiple‐drug resistance develops against well‐tolerated drugs. Plasmodium falciparum drug‐resistant malaria originates from chromosome mutations. We believe that our results, besides shedding light on the mechanism of chloroquine resistance in P. falciparum, have implications for the development of novel therapies against resistant malaria strains and demonstrate the usefulness of an approach combining systems biology strategies with structural bioinformatics and experimental data. This review summarizes the commonly used antimalarial drugs, their mechanism of action and the genetic markers validated so far for the detection of drug-resistant parasites. Compare Plaquenil vs Chloroquine - Plaquenil hydroxychloroquine has been shown to improve physical function in up to 80% of patients with rheumatoid arthritis. Plaquenil hydroxychloroquine is commonly used for lupus in children because of less side effects compared to other medications. Plaquenil hydroxychloroquine helps minimize joint damage.
